The OLIS DSM acquisition achieves zero drift, near-perfect baseline flatness, and outstanding
S/N quickly. Spectral data collection rates start at 0.1 second per datum during scanning and
kinetic data collection rates are up to 1,000 points per second. All electronics and software are
optimized for sensitivity and speed, including a 16 bit, 500 kHz analog-to-digital card. Fully
digital design means that all sorting, summing, ratioing, logarithms, noise filtering, et cetera, of
raw digital data are done by the OLIS CD SpectralWorks program, our fully Windows 7 Pro
compatible software for providing maximum precision in operation and data handling.
The sample compartment is the "Polarization Toolbox," which uniquely offers the freedom to
position the polarizers and photoelastic modular not only for CD but in those positions
supporting Circularly Polarized Luminescence and Polarization of Fluorescence. These
secondary measurements support study of the molecules in their excited state using an
electro-optical method which acquires L-format polarization of fluorescence and anisotropy
directly without G-factor corrections, as well as fluorescence detected circular dichroism
(FDCD) and CPL.
The optical bench is the premium prism-grating double monochromator by CARY with an
available spectral range of 185-2600 nm fitted with a 150 watt xenon arc lamp, regulated
power supply and ignitor. For UV CD of protein, peptide, nucleic acid studies, the spectral
data are acquired with the best detector extent: a UV optimized photomultiplier tube. For NIR
scanning (800-1700), the fast response, low noise InGaAs detector can be purchased and
added.
The emission wavelength selector is a compact single grating monochromator to support
scanning emission work and for ease of selection of any appropriate emission wavelength.
This monochromator can be set to zero, and thereby act as a mirror. It can be for any emission
from approximately 300-800 nm with or without associated scanning. Should higher stray
light rejection ever be required, a filter can be inserted in the filter holder, producing a double
monochromator.
The detector for emission (e.g., FDCD and CPL) is a high sensitivity gated photon counting
detector with spectral range of 230-850 nm. The counts streaming from the photon counting
photomultiplier are alternately routed to a 'left' counter and a 'right' counter in synchrony and
in appropriate phase relationship to the operation of the PEM in the analyzer's optical train.
Left and right counts are accumulated until a useful signal-to-noise ratio has been achieved.
CPL data acquisition can last hours and longer, so the absolute stability of the Olis method
greatly enhances the likelihood of successful results.
The software is a comprehensive data acquisition/analysis/instrument control program which is
easy to use and dense with useful functions. Everything for 2D and 3D data acquisition and
analysis is included. After spectra are acquired, they can be saved as binary, ASCII, Excel, or
JPG. Analysis options include three popular algorithms for secondary structure determination
(SELCON3, CONTINLL, and VARSLC) and an exhaustive selection of 2D data manipulation
choices (smoothing, derivatives, exponentials, subtraction, etc.) as well as kinetic fits. Three
dimensional data acquired during thermal melts, chemical denaturation, and kinetic studies are
analyzed with Singular Value Decomposition (SVD) to determine the number of species in the
reaction, followed by Downhill Simplex and Matrix Exponentiation for spectral reconstruction
of the starting, final, and any spectral intermediate forms.
