The OLIS DSM 17 CD collects circular dichroism by definition: CD = abs(L) - abs(R). This is
achieved by our "Digital Subtractive Method" (DSM), which uses a 16 bit analog-to-digital
converter and proprietary software to lock-into the 50 KHz modulation rate of the PEM and
collect the raw signal, i.e., noise plus sine wave, therefore AC and DC. This is a markedly
superior alternative to the 50 year old method of calculating CD as the ratio of decoupled AC
and DC intensities times a calibration constant.
In DSM, every datum -- 10 points per sine -- is tagged as abs(L) or abs(R). Consequences of
this modern digital method include (1) no decoupling of AC and DC information, (2) no need
for the calibration required by analog lock-in amplifier methods, (3) no electronic drift, (4)
linearity over at least 7 orders of magnitude, (5) raw data for reversible processing, which also
results in simultaneous acquisition and (optional) presentation of absorbance spectra with
the CD.
Built around conventional-speed scanning prism and grating monochromator with spectral
range of 185-2600nm; detection range is fixed by the lamp and detectors to 185-1700 nm. For
this detection range a 150 watt ozone producing lamp (150 W non ozone producing lamp can
be requested if wavelengths below 220 nm are not needed) is used. The xenon arc lamp is used
with a closed cycle cooling system and shut-off safety feature, guaranteeing against the lamp
overheating; use of the lamp is recorded on the lamp power supply. The detectors used for the
185-1700 nm range are PMTs and InGaAs detectors.
Tagging, sorting, summing, ratioing, logarithms, all noise filtering, et cetera, are handled by
the OLIS SpectralWorks program, our software for optimizing the user experience and
handling the workload of analog hardware still used by other CD manufacturers.
This SpectralWorks is a comprehensive data acquisition and instrument control software
package, which also includes algorithms for secondary structure determination, 3D data
analysis, and an exhaustive selection of 2D data manipulation choices (smoothing, derivatives,
exponentials, subtraction, etc.).
The provided computer hardware includes an OLIS verified and supported desktop PC with
3rd Gen. Intel Core i3-3240 3.4GHz, 4GB DDR3, 1TB HDD 7,200RPM, DVD-RW drive, 6
Total USB 2.0 Ports. Windows 7 Pro 64-bit (English version required).
Client to furnish two volt AC outlets and fast flow nitrogen source.
Quartz cuvettes are available for purchase separately.
