Research quality scanning spectrofluorimeter with one OLIS double grating f/4.2 Hummingbird monochromator on the excitation side and one OLIS single grating "sister" monochromator on the emission side. Excitation source is a water-cooled 150 watt xenon arc lamp run within a mirrored elliptical lamp housing designed for maximum intensity. The lamp is ignited and run with a high precision regulated power supply for highly reliable lamp starting, ultralow ripple, and high precision current regulation. Computerized emission and excitation wavelength selection is supported, with software settable scanning and signal averaging. Detection rates are up to 10 milliseconds, with an arbitrary amount of signal averaging during or after collection to optimize for speed or best sensitivity. One digital photon counting module (PCM) is provided for exquisite sensitive emission readings from 230-870 nm. A second detector, a high precision analog photomultiplier tube, is used for the absorbance readings, mounted directly adjacent to the sample.
The sample compartment is a single beam CLARiTY Chamber. The "CLARiTY" chamber eliminates the effect of scatter and thus allows accurate absorbance and fluorescence spectroscopy to be done on turbid sample suspensions. Within the "CLARiTY" chamber is the 'suspension presentation cavities,' (DSPC) which is either a high-absorbance (up to 22 AU) micro cuvette, or a 1mL, or 8 mL (low absorbance, to nanomolar concentration) quartz sample holder . The DSPCs return effective pathlengths of 0.25 millimeters, 5 centimeters, or 30 centimeters respectively. A large DSPC is best when sample volume is of no concern, as when it is E.coli, yeast, algae, and all other plentiful and/or inexpensive samples are used. The benefit of the large volume is the tremendous pathlength enhancement of approximately 30 cm. The small DSPC allows one to work with finite sample volume (e.g., embryonic zebra fish or infant blood cells). The pathlength enhancement is far less, only 4 centimeters for a 1 mL volume. But, one is flooding the smaller volume with the same light intensity, so there is a stronger detected signal and thus superior S/N. Ports holding quartz rods are built-in, allowing light in and out of the otherwise sealed DSPC. The sample itself is introduced through the top of the DSPC manually (frequently, using a pipette) or with the OLIS Automatic Titrator(quoted separately).
In addition, a water jacketed cell holder for a normal 1 cm2 cuvette is included. This opens the use of the spectrophotometer/ fluorimeter for those times when the pathlength enhancement and/or scatter immunity of the DSPC is not needed or appropriate. The two sample holder assembles are quickly exchanged one for the other. Order water bath separately.
The Windoes 10 compatible SpectralWorks software is provided for all instrument control, data acquisition, data handling, and data fitting. Fitting algorithms include those of the Matheson-DeSa GlobalWorks suite, plus many for 2D analysis including peak finder, spectral deconvolution, difference spectra, derivative, integration, etc. Data storage and retrieval, with import/ export capabilities for ASCII and EXCEL file types are provided for dealing with OLIS and non-OLIS acquired data. The provided computer hardware is an OLIS verified and supported desktop PC with 3rd Gen. Intel Core i3-3240 3.4GHz, 4GB DDR3, 1TB HDD 7,200RPM, DVD-RW drive, 6 Total USB 2.0 Ports. Windows 10 Pro 64-bit (English version required), Keyboard & Mouse and 20" Class Monitor.
Software supports data acquisition and handling modes for scanning excitation, fixed wavelength excitation, scanning emission, fixed wavelength emission, synchronous scanning of emission and excitation, light scatter, and single beam absorbance of data in 2D and 3D. Base of the instrument dimensions are approximately 22" x 29".
