The sample compartment is the "Polarization Toolbox," where the polarizers and photoelastic modulator are positioned to support Circularly Polarized Luminescence.
The excitation light source, is a wavelength specific LED, filtered to reduce the bandwidth of the produced color to +/-5nm. OLIS electronics hold the intensity stable to +/-0.001 volts/24 hour cycle. The filtered and LED is housed in the excitation tube designed for easy interchange. LEDs in the 400nm-1100nm range are considered are standard but UV wavelengths in the 240-390nm region are available at higher price. One LED Excitation Tube is included with the price of the instrument. Additional Excitation Tubes are quoted separately, below. A NIR monochromator and NIR detector is available at higher cost.
The emission wavelength selector is a compact single grating monochromator to support scanning emission work and for ease of selection of any appropriate emission wavelength. This monochromator can be set to zero, and thereby act as a mirror. It can be for any emission from approx imately 300-800 nm with or without associated scanning. Should higher stray light rejection ever be required, a filter can be inserted in the filter holder, producing a double monochromator.
The detector for emission is a high sensitivity gated photon counting detector with spectral range of 230-850 nm. The counts streaming from the photon counting photomultiplier are alternately routed to a 'left' counter and a 'right' counter in synchrony and in a ppropriate phase relationship to the operation of the PEM in the analyzer's optical train. Left and right counts are accumulated until a useful signal-to-noise ratio has been achieved. CPL data acquisition can last hours and longer, so the absolute stability of the Olis method greatly enhances the likelihood of successful results.
The software is a comprehensive data acquisition/analysis/instrument control program which is easy to use and dense with useful functions. Everything for 2D and 3D data acquisition and analysis is included. After spectra area cquired, they can be saved as binary, ASCII, Excel, or JPG. Analysis options include three popular algorithms for secondary structure determination (SELCON3, CONTINLL, and VARSLC) and an exhaustive selection of 2D data manipulation choices (smoothing, derivatives, exponentials, subtraction, etc.) as well as kinetic fits. Three dimensional data acquired during thermal melts, chemical denaturation, and kinetic studies are analyzed with Singular Value Decomposition (SVD) to determine the number of species in the reaction, followed by Downhill Simplex and Matrix Exponentiation for spectral reconstruction of the starting, final, and any spectral intermediate forms.
